De collectie bestaat uit ongeveer zesduizend stukken van oude en moderne kunst, waarvan slechts duizend en dertig worden blootgesteld aan het publiek.
Het museum opende zijn deuren in oktober 1969, in navolging van de testamentaire bepalingen van Calouste Sarkis Gulbenkian, een industrieel van Armeense afkomst, die halverwege de twintigste eeuw in Portugal was gevestigd en die zijn hele leven lang een uitgebreide kunstcollectie samenbracht. Het gebouw waar het museum zich bevindt, gelegen in het Santa Gertrudes Park, is geschreven door Alberto Pessoa, Pedro Cid en Ruy de Athouguia.
De permanente tentoonstellingsstukken worden zo belicht dat ze twee onafhankelijke circuits vormen. De eerste is gewijd aan oosterse en klassieke kunst, met stukken Egyptische, Grieks-Romeinse, islamitische kunst, kunst uit China en kunst uit Japan.
Het andere circuit is gewijd aan Europese kunst, met stukken die chronologisch zijn gelokaliseerd in een periode die begint in de elfde eeuw en eindigt in de twintigste eeuw. Dit circuit heeft kernen gewijd aan boekkunst, decoratieve kunst, beeldhouwkunst en schilderkunst. Wat deze laatste kern betreft, moeten de werken van kunstenaars als Domenico Ghirlandaio, Degas, Édouard Manet, Peter Paul Rubens of William Turner worden benadrukt.
